Transgene
description
Transgene Type:    Transgenic (Inserted expressed sequence)
Mutation:    Insertion

Tg(NR2E1)10Ems expresses 1 gene

Mutation detailsThe transgene comprises the 141 kb clone dJ429G5 from the human genomic library RP3, which spans NR2E1. RT-PCR with human-specific primers demonstrated mRNA expression in whole adult brain. (J:99431)

Notes
Presence of the transgene rescues the phenotype of mice homozygous for the fierce (Nr2e1frc) mutation. Both gross and microscopic structural abnormalities of the brain are corrected in these mice, as are the radial asymmetry and mottling of the retinal pigment epithelium of the eye; the number of retinal vessels increases, though not to wild-type levels. The fierce behavioral phenotype, including high spontaneous activity level, large proportion of time spent in and force of struggling when lifted by the tail, high aggression level in the resident-intruder test, and small proportion of time spent in social interaction, is completely abrogated by the transgene. Whereas male fierce mice will attack and kill females, rather than mate with them, these mice exhibit no aggression toward females and mate successfully.

Five independent lines were established from 8 potential founders. Lines 10 and 11 were selected for characterization. No differences were observed between the two lines for mice of any genotype, whether only transgenic or in combination with the fierce mutation, so data for the two of them were pooled.
