Mutation detailsA targeting vector was designed to insert a T1799A mutation into exon 15 and a loxP-stop-loxP cassette containing a minigene encoding exons 15-18 of wild-type Braf with a splice acceptor sequence at the 5' end into intron 14. Upon cre expression, the loxP-stop-loxP is deleted, enabling expression of the knock-in sequence. This mutation increases the kinase activity of the protein. (J:104375)
