Immunohistochemical analysis does not detect ovalbumin (OV) in the pancreatic islet beta cells of mice bearing this transgene, nor does western blot analysis, indicating that their OV content is less than 0.03 ng/ug of protein. Mice expressing both this transgene and Tg(Tcra,Tcrb)1100Mjb (OT-1), which encodes an ovalbumin-specific T cell receptor expressed on CD8+ T lymphocytes, develop early onset, spontaneous diabetes with islet infiltration, implying that the beta cells express OV. While only one of 60 lethally irradiated Tg(Ins2-OV)307Wehi mice receiving a 1:4 (OT-1 mouse:C57BL/6-Thy1a congenic) bone marrow mixture became diabetic during the 10 weeks post transfer, histological evaluation of these mice at 15-21 weeks post-transfer showed mild infiltration of 22% of their islets.
