Mutation detailsThe coding sequence of the transgene consists of the puromycin acetyltransferase coding sequence fused in-frame to a 5' truncated HSV1 thymidine kinase gene that begins at the second ATG of the full-length gene and encodes a protein lacking the carboxy-terminal 45 amino acids. This construct resides upstream of the bovine growth hormone polyadenylation signal and downstream of the phosphoglycerate kinase 1 promoter followed by a loxP-flanked neomycin acetyltransferase gene with the bovine growth hormone poly-A signal. The transgene was inserted in reverse orientation into exon 3 of the endogenous Hprt gene via homologous recombination. (J:99258, J:99558)

Notes
When mice bearing this transgene are crossed with mice transgenic for cre, excision of the loxP-flanked neo gene in cells expressing Cre recombinase results in expression of a puromycin acetyltransferase-thymidine kinase fusion protein under control of the (ubiquitously-expressing) Pgk1 promoter. Treatment of the mice with ganciclovir (GCV) causes specific ablation of cells expressing HSV1 Tk activity.
