Mutation detailsThe Jun oncogene cDNA was inserted into the L7deltaAUG vector, which is a PCR-mutated Purkinje cell protein 2 gene. The vector comprises the entire genomic sequence of Pcp2, including 1 kilobase of 5'-flanking sequence and about 200 bp of 3'-flanking sequence, modified to eliminate a BamHI restriction site in the third intron and all potential transcription start sites. The cDNA was inserted into the only remaining BamHI site, in exon 4, so transcription occurs from the Pcp2 promoter and translation from the initiation site of the cDNA. (J:96313, J:96314)
