Transgene
description
Transgene Type:    Transgenic (Inserted expressed sequence)
Mutation:    Insertion

Tg(Th-SNCA*A53T)1Ddim expresses 1 gene

Tg(Th-SNCA*A53T)1Ddim expression driven by 1 gene

Mutation detailsThe transgene contains 4.8 kb of rat tyrosine hydroxylase (Thy1) promoter sequence, a human alpha synuclein (SNCA) cDNA sequence carrying a G209A transition mutation, which encodes an Ala53Thr amino acid substitution, and an SV40 polyadenylation sequence. (J:70209)

Notes
Six transgenic lines were generated from the A53T construct. The transgene is expressed within nigrostriatal dopaminergic neurons. A similar and consistent pattern of transgene expression in all lines of transgenic mice was revealed by in situ hybridization studies. Transgenic protein accumulation impacted neither the number of nigral neurons, nor the levels of striatal dopamine in animals up to one year of age. No overt nuerodegerative changes were detected in transgenic mice.
