Transgene
description
Transgene Type:    Transgenic (Inserted expressed sequence)
Mutation:    Insertion

Tg(ACTB-Ica1/EGFP)18Mdos expresses 1 gene

Mutation detailsA chicken beta actin promoter drives the expression of the fusion gene Ica1/EGFP, which encodes an islet cell autoantigen 1 (Ica1) molecule in which a full-length enhanced green florescent protein (EGFP) replaces the Ica1 natural 3'-stop codon. Tissue fluorescence is strongest in the heart, intermediate in the exocrine pancreas and kidney and weak in the liver. Tissue fluorescence is observed in isolated transgenic islets. No fluorescence was observed in the brain, thymocytes or spleen cells.

Notes
Line 18 transgenic mice are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. NOD transgenic females exhibit reduced diabetes incidence (45%) by 40 weeks of age and more rapid insulitis onset (50%, severe infiltration) at 12 weeks of age compared with NOD controls (80% diabetic by 40 wk and 20% insulitis-positive at 12 weeks). Intra-islet lymphocytes expressing interferon gamma were 50 times more frequent in sections from transgenic Line 18 than in wild-type controls, while IL4 stained cells were barely detectable in either.
