Mutation details: The transgene comprises the entire coding sequence of human cytochrome P450, family 4, subfamily b, polypeptide 1 (CYP4B1) cDNA coupled to the 5' flanking region and hepatic control sequence of the human apolipoprotein E gene. Immunoblot analysis of hepatic microsomes from transgenic mice revealed an immunoreactive protein having the mobility of human CYP4B1. Lauric acid omega-hydroxylation was 25% greater and mutagenic activation of 2-aminofluorene two-fold greater in transgenic than in control liver microsomes; both these activities were abrogated by antibodies to CYP4B1.
