Mutation detailsA 0.6-kb genomic DNA fragment containing exon 2, including the translation initiation codon, exon 3, and part of exon 4 was replaced with a cassette containing a puromycin resistance gene, a loxP site oriented distally (away from the centromere), the 3' half of an Hprt minigene, and a tyrosinase (Tyr) minigene in an ES cell line already harboring Wnt3tm2Brd. The cassette integrated into the same Chromosome 16 homolog, in cis with Wnt3tm2Brd. (J:56548, J:80303)

Oppositely-oriented loxP sites in Trp53tm2Brd and Wnt3tm2Brd, in cis, served as the proximal and distal breakpoints, respectively, of a 24-cM Cre recombinase-mediated inversion, In(11Trp53;11Wnt3)8Brd. (J:56548) Passage of the two targeted mutations through the germline without exposure to Cre recombinase resulted in their segregation to separate Chr 16 homologs and thus to their independent inheritance. (J:80303 )
