Mutation detailsThe gene was disrupted by replacement of exons 5 and 6 with a neomycin resistance cassette via homologous recombination, resulting in a frameshift and premature translation termination in exon 7. Gene expression was absent in homozygous mutant animals as demonstrated by Western blot analysis of samples prepared from brown adipose tissue. The same samples were assayed for enzyme activity. Samples taken from homozygous mice exhibited negligible amounts of enzyme activity. (J:78828)
