Mutation detailsInsertion of a hygromycin resistance cassette into one allele of the Rb1 gene replaced part of intron 3 and introduced three nucleotide changes into exon 3, creating two termination codons and a new PstI site. This was accomplished in ES cells in which Rb1 had already been targeted. The resulting ES cells are Rb1tm1Tyj/Rb1tm2Tyj. (J:59268)

Homozygous mutants at this locus are embryonic lethal. ES cells homozygous for functional knockout of this locus were created so that phenotypes resulting from homozygous cells could be studied in adult chimeras. The contribution of double knockout cells to the composition of chimeric mice varied from 30 to 60%.
