Name: prostaglandin-endoperoxide synthase 2; targeted mutation 1, University of North Carolina
MGI ID: MGI:2158457
Synonyms: Cox2-, COX-2-, PGHS-2-, Ptgs2tm1Smi
Gene: Ptgs2  Location: Chr1:150100031-150108227 bp, + strand  Genetic Position: Chr1, 63.84 cM, cytoband H1
Germline Transmission:  Earliest citation of germline transmission: J:29510
Parent Cell Line:  BK4 (ES Cell)
Strain of Origin:  129P2/OlaHsd
Allele Type:    Targeted (Null/knockout)
Mutations:    Insertion, Intragenic deletion
Mutation detailsA neomycin selection cassette was inserted into exon 8. A 104 bp DNA fragment containing sequences encoding essential residues tyrosine 371 and histidine 374 was also deleted. Northern blot analysis on mRNA derived from LPS-induced macrophages from homozygous mice demonstrated that an aberrant 6.0 kb transcript is produced from this allele. However, western blot analysis confirmed that no detectable protein is produced from this allele in LPS-induced macrophages derived from homozygous mice. (J:29510)
