Mapping and Phenotype information for this QTL, its variants and associated markers

J:54925

The authors examined the gentic basis of differences in seizure behaviour using a GABAa receptor agoinst beta CCM (methyl beta-carboline-3 carboxylate). A/J mice are more susceptible to beta-CCM induced myoclonic seizures than C57BL/6J mice in both latency to seizure and proportion of tested mice that seized. A genome wide screen in an F2 population of these parental strains identified a locus on proximal Chromosome 7, near D7Mit308, that displayed an association with seizure susceptibility (chi square =14.8; p = 0.006; n = 267) and seizure latency. This locus was named Bis4 with a LOD score = 3.71 and explaining ~ 22% of the genetic variance and 5-7% of the phenotypic variance for this trait. The best-fitting logistic regression model suggests that the A/J allele and the D7Mit308 locus increases the likelihood of seizures at ~3.3 fold. Candidate genes in this region include Atp1a3, Atp1b3-rs2, Grik5, Scn1b, Gabra5, Gabrb3, Gabrg3 and Szf1.
