Mutation detailsThe targeting vector was sequentially assembled using BAC clone RP24-335G16 and RP23-412J16 from the C57BL/6 library with recombination sites and the Neo cassette selection cassette flanked by SDA (self-deletion anchor) sites. The TAA stop codon in exon 9 of the hypoxanthine phosphoribosyltransferase 1 (Hprt1) gene has been replaced with a T2A-CreERT2- P2A-EGFP cassette. A T2A self-cleaving peptide sequence was placed directly downstream of the Hprt1 coding sequence to separate Hprt1 and the CreER fusion protein. The subsequent P2A self-cleaving peptide separates the Hprt1 and CreER coding sequences. The neo cassette was removed by SDA. (J:101977)
