Mutation detailsThe transgene contains the following: the Pomc gene, including sequence 8 kb downstream of the transcription start site and 13 kb upstream (minus the -6.5 to -0.8 kb region), containing pituitary-specific enhancer Rr281 but with neuron-specific enhancer Rr279 (nPE1) deleted (585 bp) and Rr280 (nPE2) still present, and the promoter, with the EGFP reporter gene and SV40 poly(A) signal sequences inserted upstream of the translation start site. The nPE2 enhancer sequence was modified with six homeodomain binding sites (HDBSs) mutated from TAAT, ATTA, TCAAG, TAAT, ATTA and TCAAT to TGGT, ACCA, TCCCG, TGGT, ACCA and TCCCT. (J:224010)
