Mutation details: A 186 kb bacterial artificial chromosome (BAC) clone RP23-157J4, spanning from Chr1:172,236,604-172,422,466 (GRCm38/mm10) was used to obtain mouse genomic DNA containing the entire mouse potassium inwardly-rectifying channel, subfamily J, member 10 (Kcnj10) promoter and regulatory sequences. This BAC was modified to replace the Kcnj10 coding sequence (genomic region corresponding to NM_001039484.1: exon 2 nucleotide 243-1393) with a ZsGreen expression cassette. The BAC contains numerous other genes in addition to Kcnj10, including Atp1a4 (partial), Igsf8, Atp1a2, Kcnj9, and Pigm which were not disabled or modified on the BAC Tg. The linearized BAC was microinjected into fertilized oocytes derived from breeding B6SJLF1/J mice together. Four transgenic founder mice were obtained by random integration. A founder line 858 with a similar ZsGreen expression pattern was selected. Line 858 has 2 copies of the transgene integrated into chromosome 15 between positions chr15:41002911 and chr15: 41067635 in intron 5 of the zinc finger protein, multitype 2 (Zfpm2) gene, resulting in a 64,724 bp deletion in intron 5 of Zfpm2 between the insertion sites.
