Mutation detailsTHe transgenic construct contains (from 5' to 3') a CAGG (CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid) promoter, three pairs of heterospecific lox sites (loxN, lox2272 and loxN), and the UFObow construct. The three pairs of heterospecific Lox sites are inserted separating a luciferase and three blue-excitable fluorescent proteins: highly bright secreted luciferase (secNluc), fast-maturating cyan fluorescent protein (mTFP2b), Large-Stokes-shift membrane-bound orange fluorescent protein (LSSmKOb), and Large-Stokes-shift far-red fluorescent protein (LSSfRFP). secNluc is constitutively expressed in all cells. To ensure that only one fluorescent protein is expressed at any one time, an SV40 polyadenylation signal and a WPRE sequence were inserted downstream of each open reading frame. Upon cre activation, the lox sites recombine to create three mutually exclusive excision possibilities with the fluorescent protein that is placed first after the CAG promoter being expressed.
