Mutation details: The transgene contains the following elements: Igf2 promoter 3 (Rr146; from 162 bp upstream of TSS of exon 3/alternative exon 1 [chr7:142211218 GRCm39] to 74 bp of 5' UTR in exon 3), an Igf2 enhancer (Rr418) from between H19 and Igf2, the luciferase reporter gene and SV40 poly(A) signal sequences. A number of independent lines were generated; this allele replaced ~15.9 kb sequence upstream of Sox10 that contains an enhancer (Rr505561), leading to disruption of its expression when homozygous. The Igf2 enhancer drives expression of the reporter in the brain.
