Mutation details: The transgene insert contains the following elements: an FRT site flanked neomycin resistance gene cassette, Gata3 promoter 1b (-1314 to +1 relative to ATG start site; contains downstream promoter, alternative 1st exon 1b, intron 1b and 5' UTR part of exon 2), the EGFP reporter gene cDNA, and BAC RP23-43G17 (which contains Gata3 T cell and thymic NK cell enhancer (7.1 kb KpnI-SalI T cell element) Rr338) where Rr338 was replaced with an FRT site flanked neomycin resistance gene cassette. The neo cassettes were removed through subsequent flp-mediated recombination. EGFP expression is not observed in thymocytes anymore.
