Mutation detailsThe transgene insert comprises a modified BAC RP23-61C14 (containing the 3' end of Myh3, Rr194, Myh2, Myh1, Myh4 and the 5' end of Myh8) where the following modifications were made: a vector containing the YFP reporter gene, a poly(A) signal cassette and a loxP site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh2 gene; a vector containing the tdTomato reporter gene, a poly(A) signal cassette and a lox2272 site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh1 gene; a vector containing the CFP reporter gene, a poly(A) signal cassette and a loxN site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh4 gene. The kana cassettes were removed through subsequent cre-mediated recombination. Super enhancer Rr194 controls expression of the Myh genes. Two copies of this transgene are integrated into the genome.
