Mutation details: The transgene insert comprises part of a modified BAC RP23-61C14 (containing Myh2, Myh1 and the 5' end of Myh4) where the following modifications were made: a vector containing the YFP reporter gene, a poly(A) signal cassette and a loxP site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh2 gene; a vector containing the tdTomato reporter gene, a poly(A) signal cassette and a lox2272 site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh1 gene; a vector containing the CFP reporter gene, a poly(A) signal cassette and a loxN site flanked kanamycin resistance gene cassette was inserted at the ATG start site of the Myh4 gene. The kana cassettes were removed through subsequent cre-mediated recombination. This transgene lacks super enhancer Rr194, which controls expression of the Myh genes, in contrast with Tg(Rr194,Myh2-YFP,Myh1-tdTomato,Myh4-CFP)Enh+Mair.
