The targeting cassette is composed of En2 intron/splice-acceptor/exon, an IRES, the beta-galactosidase (LacZ) gene, a polyadenylation signal (pA) a loxP site, a beta-actin promoter, the neomycin-resistance gene (neo), a second pA site, second FRT site, and second loxP site. A third loxP site was inserted downstream of exon 3. In situ hybridization confirmed absence of mRNA in homozygotes. (J:320628)
