Mutation details: The transgene insert consists of BAC RP24-238B22, containing the Casp8 gene, which was modified as follows: phage T7 epitope tag coding sequence was inserted upstream of the Casp8 stop codon and an FRT site, an IRES, the GFP green fluorescent protein gene and a second FRT site, were inserted downstream of the stop codon. A twenty nucleotide sequence in intron 1 was replaced with a different sequence of equal length and composition to improve genotyping. Aspartic acid codon 387 was mutated to an alanine codon (p.D387A) to create a noncleavable version of the encoded peptide.
