Mutation details: The allele contains four chicken beta-globin HS4 insulator sequences (two on each ends) flanking the ubiquitous and strong CAG promoter driving LoxP-flanked (floxed) DNA sequence which has the N-PhiM reporter gene encoding a noncytotoxicand non-fluorescent mutated PhiYFP protein with a nuclear localization signal and a transcription stop signal followed by bright membrane-localized fluorescent reporter (tdTomato*) and cre gene separated by 2A cleavage site (T2A; mediates ribosomal skipping), then a WPRE sequence, an SV40 polyA signal. A Frt flanked PGK-neo cassette followed those sequences. Despite lack of recombination/deletion of the floxed cassette by Cre, the modified dtTomato and Cre protein are constitutively active and permanently expressed.
