Mutation detailsEnhancer sequence from intron 3 of the human PMP22 (peripheral myelin protein 22) gene was inserted upstream of the mouse heat shock protein 1a promoter driving the expression of a luciferase gene extracted from the pGL4 vector. This enhancer was called the +11 enhancer for its location approximately 11kb downstream of the translation start site (chr17:15,090,965-15,092,611). It contains, in order, two Egr2, two Sox10 and two more Egr2 binding sites. The inserted fragment contains all except the first Egr2 binding site. The mouse promoter sequence also contains the first seven Hspa1a coding nucleotides and also contains Hspa1l promoter sequence (or serves as a bidirectional promoter) and Hspa1l 5' UTR sequence. Two mouse lines, D and E with transgene copy numbers of 6 and 9 respectively, were used for further experiments. (J:274686)
