Mutation detailsThe transgene comprises the following elements: the mouse Myh6 promoter, sequence for the human Myc epitope tag immediately after the ATG start codon, the mouse Mybpc3 cDNA with a deletion of the LAGAGRRTS codons (282-290 in SW:Q3UIK0) (the equivalent on peptide level of deletion of TSLAGAGRR (280-288)), and human GH gene poly(A) signal sequence. The deletion of the nine amino-acids in the encoded peptide prevents phosphorylation by protein kinase A and proteolysis by calpain. Three mouse lines (156, 157, 165) were established; line 156 was the highest expressing line (approximately 3.5x endogenous mRNA level) and was used for further experiments. (J:133394)
