Mutation details: A targeting vector containing IRES-cre coding sequence followed by a polyadenylation signal sequence, and a FRT site flanked PGK-neo cassette was inserted immediately following the stop codon in exon 21 of the targeted Kit gene. The construct was electroporated into embryonic stem (ES) cells. Chimeric mice were generated by aggregating correctly targeted ES cells by tetraploid complementation. The resulting chimeric animals were tested for germline transmission.
