Mutation details: The transgenic construct contains a rat enolase 2, gamma neuronal (Eno2) promoter, containing a random filler sequence and an SV40 polyadenylation signal, directing expression of human matrix metallopeptidase 9 (MMP9) cDNA. The MMP9 gene was modified to harbor a glycine to leucine mutation at residue 100 (MMP9*G100L), in the cysteine switch autoinhibitor region, rendering the enzyme constitutively active. Of the five lines, founder line exhibits the highest transgene activity.
