Mutation details: The transgenic construct was derived from that used for Tg(RP23-159N6*)43Bdm, in which the sequence encoding the zinc finger domain of Prdm9 in the C57BL/6J-derived mouse bacterial artificial chromosome (BAC) RP23-159N6 was replaced by the corresponding segment from the Prdm9wm7 allele of strain B10.A(R209)-H2aw9. The present transgene has been further modified by mutation of codons 357-358 from TATGGG to TTTGGT to give rise to a mutant PRDM9 protein in which tyrosine has been replaced by phenylalanine at amino acid position 357 (Y357F).
