Mutation details: This 'knockout first' allele was created using the KOMP targeting vector for this gene (PRPGS00082_A_A10). It inserts a beta galactosidase (lacZ) reporter gene trap and a neomycin resistance gene cassette into intron 1 and flanks exon 2 with loxP sites. The lacZ-neo double cassette is flanked by FRT sites and the two cassettes are separated by a loxP site. Insertion of the lacZ gene trap (which starts with a strong splice acceptor site) promotes splicing of exon 1 of the target gene to the lacZ gene, thus disrupting normal transcription and creating a null allele.
