Mutation details: The transgenic construct consists of a hemaglutinin (HA) epitope and the human SLC2A4 (GLUT4) COOH terminus fused to an intracellular EGFP tag under the control of a muscle creatinine kinase promoter, enhancer 1, untranslated exon 1, 3kb of intron 1 (including the enhancer 2 region), and the first 16 bp of exon 2 and a poly(A) signal. The HA epitope tag is in the first extracellular loop that is exposed on the cell surface. Western blot analysis confirmed expression in skeletal muscle (soleus, gastrocnemius, extensor digitorum longus, flexor digitorum breviis). Expression is 2.2 fold higher than the endogenous protein.
