Mutation detailsA construct was generated by inserting the icre (improved cre) cDNA between the promoter region and polyA site in the IE3.9intpolyAA construct (containg the Gata1-HRD promoter (HRD; hematopoietic regulatory domain)). The transgenic vector was verified by sequencing, linearized and used for pronuclear injection. Three transgenic founders were identified from 16 chimeras, and cre activity was assessed in progeny of crossed to ROSA26 reporter animals. Reporter activation was detected only in bone marrow cells (in early erythroid progenitors) and not in any other organ or tissue examined. The line with greatest cre activity was selected for further analyses. (J:209268)
