Mutation details: The transgene contains the cardiac-specific mouse myosin, heavy polypeptide 6, cardiac muscle, alpha (Myh6) promoter through the translation initiation codon, in exon 3, joined to the coding sequence for mouse potassium voltage-gated channel, Isk-related subfamily, member 1 (Kcne1; GenBank: X60457 nt 1-611), followed by the human growth hormone polyadenylation signal. Northern blot analysis reveals markedly elevated levels of Kcne1 mRNA in heart, but not in kidney, of transgenic mice. Western blot analysis using an antibody to a C-terminal epitope of KCNE1 yields three bands representing native and glycosylated forms of the protein in extracts of hearts from transgenic, but not from wild-type, mice.
