Mutation detailsA BAC containing the podoplanin locus was modified, replacing exon 1 with a codon-optimized cre sequence and polyadenylation site. The cre sequence was integrated downstream of the start codon. Microinjection of the construct into pronuclei of zygotes produced 17 founders with 3 (406-408) positive for cre recombinase and the 5' ATG region by southern blot. These founders were designated 6, 12, and 14. When crossed to EYFP reporter mice, line 12 offspring did not have detectable cre expression, while line 6 offspring had some EYFP expression in the CD45+ve non-stromal cell compartment. Thus line 6 was excluded from further analysis. Offspring from line 14 had no cre activity in non-stromal cells and substantial activity in CD45-ve LN (lymph node) cells (in 15% in LEC and FRC fractions and very low expression in BECs or absent in the double-negative fractions.)
