Mutation detailsThe sequence of the rat NSE (neural specific enolase; Eno2) was inserted 5' upstream of cDNA encoding cre recombinase flanked by mutated mouse estrogen receptor ligand-binding domains (MerCreMer). The construct was injected into fertilized oocytes and 6 founders (4 male, 2 female) were generated and 2 (#'s 778 and 805) were found to show expression in the brain and intestine in F3 animals by RT-PCR or Western blot. The other 2 lines did not show protein expression in the brain or intestine despite MerCreMer transcript expression determined by RT-PCR. Transcript expression in line 778 was detected at E14.5, corresponding to neurogenesis in the CNS and ENS. Both lines were further characterized by analyses of tissue from offspring of crosses to reporter mice. Line 778 was created as a representative line. (J:187253)
