Mutation detailsCre recombinase replaced the beta-galactosidase gene in a construct containing the -742bp Col11a2 promoter sequence, SV40 RNA splice site, lacZ sequence, and Col11a2 first intron. The 4.2 kb construct was microinjected into fertilized pronuclei. 5 founders were obtained. 2 lines (1 and 5) did show recombination in crosses to lacZ reporter mice. Line 2 showed cre activity in neural tissue and line 3 had ubiquitous activity. Line 4 was chosen due to its restricted cre activity pattern, although it had a low transgene copy number compared to the other lines. (J:187063)
