Mutation detailsThe construct was constructed by inserting a cassette containing the flpe/ERT2 coding and polyA sequences into a Wnt1 expression-vector. 11 kb of Wnt1 genomic DNA containing critical 3' enhancer sequences were included after the FLPe/ERT2 cassette. Tandem insulator sequences flanked the complete construct. The purified transgene was injected into fertilized eggs to produce transgenic animals. 11 lines were obtained and FLPe/ERT2 mRNA expression recapitulated endogenous Wnt1 expression. Two lines showing the highest levels of FLPe/ERT2 mRNA were characterized with respect to recombinase activity. Data from line 9455 was shown in detail. (J:97320)
