Mutation details: Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23-395M11) containing the acid phosphatase 5, tartrate resistant (Acp5) gene. Another BAC library was used to obtain BAC #RP23-412O5, which contained the integrin binding sialoprotein (Ibsp) gene and the dentin matrix protein 1 (Dmp1) gene. These genomic fragments were linked together and different fluorescent reporters, followed by polyadenylation sequences, were inserted upstream of the translational start site of each gene. The cyan variant of green fluorescent protein (GFP) was inserted into Acp5, the topaz variant of yellow fluorescent protein (YFP) was inserted into Ibsp, and mCherry varient of red fluorescent protein (RFP) was inserted into Dmp1. Lines 1 was generated in which only the YFP reporter is functional.
