Mutation details: This transgene contains 1.1 kb of the 2.1 kb human v-ets erythroblastosis virus E26 oncogene homolog 2 (avian) (ETS2) cDNA inserted into the p1017 expression vector between the T cell-specific, proximal promoter of the mouse lymphocyte protein tyrosine kinase (Lck) gene and the human growth hormone (GH1) genomic sequence, including the GH1 polyadenylation signal. The truncated ETS2 encoded by the transgene corresponds to the DNA binding domain of the normal protein; it competes with the endogenous mouse ETS2 and thus acts as a dominant negative mutant. Based on Southern blot analysis, 20 copies of the transgene are estimated to be present. Western blot analysis of total thymocyte lysates from these mice using anti-ETS2 antibodies demonstrates expression of the truncated ETS2 protein.
