Mutation detailsA 786-nucleotide cDNA (nucleotides 34-819 of pg800) encoding the quail receptor (TVA) for avian leukosis viruses of subgroup A [ALV(A)] is expressed under the control of regulatory sequences from the mouse Kdr (kinase insert domain protein receptor) gene: a -640 bp to +299 bp genomic DNA fragment containing the promoter and a 2.3 kb fragment containing the endothelium-specific enhancer from the first intron. Embryonic day (E) 10.5 embryos of this line were shown by whole mount in situ hybridization to express TVA mRNA selectively in the developing vasculature, in a pattern similar to that of endogenous Kdr mRNA. Only endothelial cells of new vasculature induced in adult mice exhibit strong TVA immunoreactivity. (J:134089)

Proliferating endothelial cells of new vasculature induced by extracellular matrix implants or by tumors in adult mice bearing this transgene are susceptible to infection by Rous sarcoma virus derived RCAS (replication-competent, avian leukosis virus LTR, splice acceptor) avian retroviral expression vectors, demonstrated by expression specifically in these cells of green fluorescent protein from one such vector.
