Mutation details: The Thy1.2 promoter drives expression of a human dynamitin (DCTN2) cDNA, with a FLAG tag added to the 3' terminus of the coding sequences, in postnatal motor neurons. The M21 line was estimated to have a ~20-fold increase in dynamitin mRNA levels. Expression of the encoded protein in brain homogenates by western blot using FLAG-tag antibodies is not detected until postnatal day 13. Two additional lines (M5 and M13) were created and these had ~2- to 4-fold increased levels of dynamitin mRNA, respectively.
