Mutation details: This transgene was derived from a 140-kb bacterial artificial chromosome (BAC) clone containing the mouse alpha9 acetylcholine receptor gene and at least 27 kb of upstream genomic sequence. A single copy of the DNA sequence encoding a MYC tag (MEQKLISEEDLNE) was inserted immediately before the gene's termination codon, and an internal ribosomal entry site (IRES) followed by an enhanced green fluorescent protein (EGFP) cassette immediately following the stop codon. Approximately five copies of this construct were estimated to have integrated, based on comparative fluorescent in situ hybridization (FISH). Abundance in nasal epithelium of transgene-derived transcripts is approximately eight-fold that of the endogenous transcript. In addition to recapitulating the previously reported spatiotemporal inner ear expression pattern of the receptor protein, EGFP expression from the transgene exhibits dynamic changes during inner ear development.

Fluorescence in situ hybridization (FISH) analysis showed that approximately five copies of the modified BAC are co-integrated into cytogenetic band 1C4-1C5, ~36% of the distance from the pericentromeric heterochromatin-euchromatin boundary to the telomere of Chr 1. J:152365
