Mutation details: The transgene construct contained the tetracycline transactivator (tTA) coding sequence downstream of a loxP-STOP-loxP fragement. This was inserted downstream of the alpha-CamKII (Camk2a) promoter in the final construct. Pronuclei of fertilized C57BL/6 eggs were injected with the purified linearized construct. A founder line 1 was established by backcrossing the founder with C57BL/6 mice.
