Mutation detailsA mouse BAC (clone RP23-359O19, size 222 kb) carrying the intact elastase I (Cela1) gene was used. The BAC was modified to express CreERT. For recombination-mediated targeting, arms homologous to the sequence of elastase I gene between the last exon and the polyA signal (in lower case) were added to either side of the IRES-Cre-ERT-frt-Kan-frt cassette by PCR. Using homologous recombination, the IRES-Cre-ERT-frt-Kan-frt cassette was inserted into the elastase I gene. The Kan selection marker was then removed by flp-mediated recombination. two BAC-Cela1-CreERT founders were identified by genotyping. Q-PCR based analysis indicated the presence of 2-3 copies of the transgene in BAC-Cela1-CreERT founders. (J:148748)
