Mutation details: The construct encodes a bidirectional reporter transgene. The cDNA of the chemokine KC (Cxcl1, from +2 to +140) was cloned into the pBI-G vector (Clontech). The vector contains a tetracycline responsive element consisting of seven copies of the tet-operator (tetO) that is between 2 CMV minimal promoters which are silent in absence of an activator (tTA or rtTA) sequence. When activated, one promoter drives expression of Cxcl1 while the other promoter drives expression of a beta-galactosidase reporter gene.
