Mutation details: The bacterial artificial chromosome (BAC) clone RP23-401D9 containing the mouse Gt(ROSA)26Sor gene (and 153 kb of upstream and 15 kb of downstream sequence) was modified by inserting a red-shifted variant Enhanced Green Fluorescent Protein (EGFP; Clontech No. 6029-1) cDNA and polyadenylation sequence at nucleotide 521 of the first exon of transcript 1. Founders with a single copy of the transgene (founder line I1) were maintained on a CD-1 background. EGFP is ubiquitously and reproducibly expressed in all tissues and organs examined. Stable and uniform EGFP expression is observed in differentiated progeny of transplanted hematopoietic stem cells (HSC) isolated from ROSA26-EGFP mice.
