Mutation details: The mouse Mdm2 splice isoform Beta (b) was placed under the transcriptional regulation of the human GFAP promoter. This Mdm2-b splice form encodes exons 2-3 and exon 12. This splice form lacks the binding domains for Trp53, Rb1, p19(ARF), Ep300(p300) as well as the nuclear localization and nuclear export signals. Mdm2-b spliceoform does contain the C-terminal zinc finger, the RING finger, and residues that are ATM phosphorylation substrates. Approximately 25 copies of the transgene were inserted into the genome. Transgene expression was highest in the brain and somewhat lower in the spleen. Low levels of expression were also detected in the liver, kidney, ovary, and testis. Expression patterns and levels were similar for all three founder lines studied in J:87962, regardless of copy number.
