Mutation detailsThe mouse Mdm2 splice isoform Beta (b) was placed under the regulation of the human GFAP promoter. This Mdm2-b splice form encodes exons 2-3 and exon 12. This splice form lacks the binding domains for Trp53, Rb1, p19(ARF), Ep300(p300) as well as the nuclear localization and nuclear export signals. The Mdm2-b spliceoform does contain the C-terminal zinc finger, the RING finger, and residues that are ATM phosphorylation substrates. Approximately 4 copies of the transgene were inserted into the genome. Transgene expression was highest in the brain and somewhat lower in the spleen. Low levels of expression were also detected in the liver, kidney, ovary, and testis. Expression distribution and levels were similar for all founder lines in J:87962, regardless of copy number.
