Mutation details: A 250 bp fragment of the mouse metallothionein promoter spanning nucleotides -185 to +65 was cloned upstream of a 4.6 kb fragment of the PML/RARA type L cDNA. The PML/RARA fusion gene included the RARA 3' untranslated region and the two presumed polyadenylation signals. 6 copies of the transgene were incorporated into the genome at a single site. ZnSo4 in the drinking water caused strong transgene induction in the liver after only 2 days of treatment (both at the mRNA and protein levels). Expression levels decreased with continuous treatment, with all expression lost by 30 days of treatment.
