Mutation details: The open reading frame was replaced with EGFP, IRES, puromycin resistance cassette, and a floxed CMV-hygro cassette. Transient cre expression removed the hygro cassette. Expression of GFP in mice bearing this allele recapitulates the highly restricted pattern of Sox1 gene expression, which occurs initially in the neural plate and thereafter is maintained in neuroepithelial cells throughout the neuroaxis until it is downregulated during neuronal and glial differentiation. GFP expression from this allele similarly mimics that of Sox1 in vitro, being undetectable in undifferentiated ES cells but becoming apparent in a significant proportion of cells upon induction of neuronal differentiation by aggregation and retinoic acid treatment.
